Description

Fusaric acid is a picolinic acid derived from the fungus Fusarium heterosporium. While it’s mechanism of action is not clearly understood,it likely inhibits dopamine beta-hydroxylase enzyme. As a mycotoxin it has negative effects on mammals, but it can be used to inhibit cell proliferation and DNA synthesis. For selection purposes fusaric acid is used to test resistance to itself in plants, allowing for the confirmation of genetic resistance to dangerous mycotoxin absorption. It is also used as a cell selective agent to differentiate tetracycline-susceptible E. coli from tetracycline-resistant. As a reagent it’s used to synthesize Bupicomide, an experimental beta blocker.

Description

Fusidic acid is a fusidane antibiotic compound that exerts potent bacteriostatic effects toward gram positive bacteria. It is commonly implemented with bactericidal antibiotics such as rifampicin. Fusidic acid is an inhibitor of protein synthesis by disrupting amino acid transfer from tRNA to ribosomes.

Fusidic acid inhibits a GTPase called elongation factor G or translocase. Impeding translocase activity hidners movement of tRNA and mRNA through ribosomes. While predominant effects are bacteriostatic, higher concentrations will result in bacterial death.

Description

Ficin is a cysteine protease enzyme which is derived from the latex of fig trees with activity similar to other cysteine protease enzymes including endopeptidases, aminopeptidases, and dipeptidyl peptidases. Ficin cleaves mouse immunoglobulin G molecules (IgG1) in the hinge region. The concentration of cysteine changes how ficin cleaves IgG1. A F(ab’)2 fragment is formed in the presence of 1mM cysteine. The F(ab’)2 fragments are smaller than whole IgG molecules and better penetrate tissue for antigen recognition while avoiding unspecific binding to Fc receptors. In the presence of 10mM cysteine, two 2 Fab fragments are produced instead. F(ab) fragments block background from primary antibody binding, and block endogenous and exposed immunoglobulins and tissues in labeling experiments.

Immobilized Ficin Agarose Resin offers a convenient solution to the production of F(ab’)2 and Fab fragments while eliminating autolysis and protease contamination and the need to remove the enzymes following digestion. Fab and F(ab’)2 can be further separated from whole IgG with either Protein A Agarose Resin (GoldBio Product # P-400) or ion exchange chromatography.

Description

L-Fucose is a six-carbon deoxy monosaccharide that is equivalent to 6-deoxy-L-galactose. L-Fucose is found on many N- and O-linked glycans present on the cell surface of a wide variety of organisms. The most well-known fucosylated glycans are the ABO blood group antigens. L-fucose present in gastrointestinal mucous represses virulence factor-encoding genes expressed by enterohemorrhagic E. coli. Mutations in the gene that encodes alpha-L-fucosidase can cause fucosidosis, leading to a buildup of fucose-containing compounds which cause cellular malfunction.

Description

ProBlock™ Gold Yeast/Fungi protease inhibitor cocktail contains optimized concentrations of various yeast and fungal protease inhibitors, which provide excellent inhibition of protease activities during protein purification from yeast or fungi. The cocktail contains both reversible and irreversible protease inhibitors to inhibit serine, cysteine, metalloproteases and specific inhibitors for yeast or fungal proteases, such as aspartic proteases. At 1X concentration, ProBlock™ Gold Yeast/Fungi inhibits over 90% of protease activities.

Some proteins require divalent cations (Ca2+, Mg2+ or Mn2+) for their biological activity and the presence of chelating agents may be detrimental to the protein’s activity. Chelators, such as EDTA, inhibit the purification of proteins by immobilized metal affinity chromatography.

ProBlock™ Gold Yeast/Fungi is supplied with additional, optional EDTA and 1,10-Phenanthroline monohydrate which are provided for optimal inhibition of metalloproteases and may be added in the extraction buffer or lysate as needed.

Description

trans-Ferulic acid is a hydroxycinnamic acid found predominantly in plant cell wall components. Given its bioavailability it has been used in several studies with bacteria such as Pseudomonas acidovorans and fungi like Sporotrichum pulverulentum for analyzing metabolism and degradation of trans-Ferulic acid. In several different studies the inclusion of trans-ferulic acid as part of a complex was useful for stabilizing or increasing the absorption of pharmaceuticals or vitamins. It has chemical properties that make it both an antioxidant and a metabolite of eukaryotes in plants. For lab applications it is used as a matrix for proteins in MALDI mass spectrometry and as a reagent for the synthesis of vanillin, along with other cyclic molecules. It can also be used to esterify methacrylated dextran and itself can form a polymerize.

Description

α-D-Fucose is a six-carbon deoxy monosaccharide. It is equivalent to 6-deoxy-D-galactose. D-fucose acts as a non-metabolizable analogue of L-arabinose and inhibits induction of the L-arabinose operon in some bacteria. Some resistant bacteria possess a mutant araC gene where D-fucose acts as a gratuitous inducer of the L-arabinose operon. D-fucose induces β-galactosidase to some extent in Lactobacillus plantarum.

α-D-fucose inhibits the eukaryotic enzyme Lysine-specific demethylase 1 (LSD1A), thereby causing gene repression. α-D-fucose is being explored in treatments for neurodegenerative diseases because of its effect on gene repression. Seen upregulated in many cancers, LSD1A overexpression pushes the cell cycle of otherwise non-transformed cells so α-D-fucose is being explored in cancers treatments as well.
