CCL3 plays an important role in the recruitment and activation of polymorphonuclear leukocytes in the acute inflammatory state, an inhibitory role in hematopoiesis, and displays inhibitory activity against strains of HIV-1, HIV-2, and SIV. CCL3 is a ligand for CCR1, CCR4, and the HIV-1 coreceptor CCR5.

CCl4 is a chemokine containing four conserved cysteine residues, of which the first two are adjacent. It is expressed and secreted by T-cells, B-cells, monocytes, and various tumor cells lines. Expression of CCL4 is induced by IL2 and mitogens, such as phytohemmagglutinin, LPS, and the peptidoglycan of S. aureus and expression is inhibited by glucocorticoids, TGF-beta, and cyclosporin A. CCL4 is an HIV-suppressive factor produced by CD8+ T-cells. It also binds to HSV-1 virions and generates pores in the envelope of HSV-1. CCL4 is a ligand for CCR1, CCR3, CCR5, CCR8, and ACKR2 (D6).

Expression of CCL5 is induced by TNF-alpha and IL1-alpha and inhibited by stimulation of T lymphocytes. CCL5 is chemotactic for T lymphocytes, eosinophils, basophils and plays a role in recruiting these cells to inflammatory sites. It also induces proliferation and activation of CHAK cells and is an HIV-suppressive factor. CCL5 is a ligand for CCR1, CCR3, CCR5, and the viroreceptor M3.

CCL7 is a chemokine containing four conserved cysteine residues, of which the first two are adjacent. It is expressed and secreted by T cells, monocytes, macrophages, and a variety of tumor cell lines. Expression is induced by IFNG and inhibited by IL13. It is chemotactic for monocytes, eosinophils, and NK cells and regulates protease secretion by macrophages which may contribute to metastasis of cancer cells. CCL7 is a ligand for CCR1, CCR2, CCR3, CCR10, and ACKR2 (D6).

CCL8 is a chemokine containing four conserved cysteine residues, of which the first two are adjacent. It is expressed by peripheral blood leukocytes, as well as a variety of other cells. Expression of CCL8 is induced by IFNG, IFNB, IL1B, and various cytokine inducers such as viruses, endotoxins, and mitogens. It is chemotactic for monocytes, lymphocytes, basophils, and eosinophils and plays a role in tumor-associated leukocyte infiltration by recruiting leukocytes to sites of inflammation. CCL8 is a ligand for CCR1, CCR2B, CCR3, CCR5, ACKR2, and ACKR4.

Ciliary neurotrophic factor (CNTF) is a polypeptide hormone whose actions appear to be restricted to the nervous system where it promotes neurotransmitter synthesis and neurite outgrowth in certain neuronal populations. It was initially identified as a trophic factor for embryonic chick ciliary parasympathetic neurons in culture. The protein is also a potent survival factor for neurons and oligodendrocytes and may be relevant in reducing tissue destruction during inflammatory attacks. In addition, CNTF is useful for treatment of motor neuron disease and it could reduce food intake without causing hunger or stress. CNTF is structurally related to IL6, IL11, LIF, and OSM. All of these four helix bundle cytokines share gp130 as a signal-transducing subunit in their receptor complexes.

Colony Stimulating Factor 1 (CSF1), previously called Macrophage Colony Stimulating Factor (M-CSF), is a hematopoietic growth factor that is involved in the proliferation, differentiation, and survival of monocytes, macrophages, and bone marrow progenitor cells. It is produced by osteoblasts (as a result of endocrine stimulation by parathyroid hormone), exerts paracrine effects on osteoclasts and can interact with CSF1R. CSF1 is a four α-helical bundle cytokine and its active form is found extracellularly as a disulfide-linked homodimer. Four transcript variants encoding three different isoforms have been reported for CSF1 gene. Although forms may vary, all of them contain the N-terminal 150 amino acid portion that is necessary and sufficient for interaction with the receptor. The first 223 amino acid of mature human CSF1 shares 88%, 86%, 81% and 74% sequence identity with corresponding regions of dog, cow, mouse and rat CSF1, respectively. Human CSF1 is active in the mouse, but mouse CSF1 is reported to be species-specific.

Colony Stimulating Factor 2 (GM-CSF) was initially characterized as a growth factor that can support the in vitro colony formation of granulocyte-macrophage progenitors. It is produced by a number of different cell types, including activated T cells, B cells, macrophages, mast cells, endothelial cells and fibroblasts, in response to cytokine or immune and inflammatory stimuli. CSF2 is also a growth factor for erythroid, megakaryocyte and eosinophil progenitors. On mature hematopoietic cells, CSF2 is a survival factor for and activates the effector functions of granulocytes, monocytes/macrophages and eosinophils. It can induce human endothelial cells to migrate and proliferate and can also stimulate the proliferation of a number of tumor cell lines, including osteogenic sarcoma, carcinoma and adenocarcinoma cell lines. CSF2 is species specific and human CSF2 has no biological effects on mouse cells.

Colony Stimulating Factor 3 (G-CSF) is a pleiotropic cytokine best known for its specific effects on the proliferation, differentiation and activation of hematopoietic cells of the neutrophilic granulocyte lineage. It is mainly produced by monocytes and macrophages upon activation by endotoxins. Various carcinoma cell lines and myeloblastic leukemia cells can express CSF3 constitutively. In humans, two distinct cDNA clones for CSF3 have been isolated. Both proteins have a 30 amino acid signal peptide and have nearly identical amino acid sequences. Human CSF3 is 73% identical at the amino acid level to murine CSF3 and the two proteins show species cross-reactivity.

CX3CL1 is a chemokine that is unlike most chemokines because it contains the C-X-X-X-C motif. It is expressed by neurons, endothelial cells, follicular dendritic cells, and some lymphocytes. Expression of CX3CL1 is induced by activation of the transcription factor NFKB1 through IFNG, IL1B, TNF, or LPS. CX3CL1 is chemotactic for T-cells and monocytes and plays a role in angiogenesis, as well as regulation of leukocyte traffic at the endothelium. It also functions as a survival factor for microglial cells. CX3CL1 is a ligand for CX3CR1 as well as virus-encoded receptors M3 and US28.

CXCL1 is an ELR type of CXC chemokine. It is secreted by human melanoma cells and expressed by neutrophils, monocytes, keratinocytes, macrophages, epithelial cells, and fibroblasts. CXCL1 participates in chemotaxis and cell activation of neutrophils. CXCR2 is the receptor that CXCL1 binds. CXCL1 inhibits embryonic oligodendrocyte precursor migration, suggesting a role in spinal cord development. It also plays a role in would healing, angiogenesis, and tumorigenesis. CXCL1 is believed to be the major inflammatory factor in recurrent herpetic stromal keratitis.

CXCL10 is a CXC chemokine lacking the ELR motif. It is expressed in and secreted by monocytes, endothelial cells, fibroblasts, and other cell types after induction by IFN-gamma. It is chemotactic for monocytes and T-cell lymphocytes. CXCL10 plays a role in angiostasis, inhibits bone marrow colony formation, and promotes T-cell adhesion to endothelial cells. Increased expression of CXCL10 and CXCR3 is also associated with autoimmune myasthenia gravis. CXCL10 also exhibits antimicrobial properties against Gram-positive and Gram-negative bacteria. It is closely related to CXCL9 and CXCL11, all three of which are ligands for CXCLR3.

CXCL13 is a CXC chemokine lacking the ELR motif. It is expressed highest in liver tissue, followed by spleen, lymph nodes, appendix, and stomach. It is a ligand for CXCR5 and is selectively chemotactic for B lymphocytes. CXCL13 plays an essential role in the development of lymph nodes and the organization of B cells within lymphoid follicles, but its expression is downregulated within lymphoid tissues by IFN-gamma during immune responses. CXCL13 induces B cells to upregulate lymphotoxin alpha-1-beta-2, leading to CXCL13 expression and creating a positive feedback loop essential for follicular dendritic cell development and homeostasis.
