GoldBio’s Goof-Proof™ Universal Probe Master Mix is a ROX-free, hot-start qPCR master mix optimized for fluorescent probe–based detection, such as SNP genotyping and quantitation. It is perfectly designed for all fluorescent probe technologies, including displacement probes such as Molecular Beacon and hydrolysis probes such as TaqMan®.
This 2X master mix delivers sensitive, high-specificity amplification with minimal background and supports multiplexing across a wide range of instrument platforms that do not require ROX normalization.
With built-in hot-start polymerase, dNTPs, and optimized buffer, Goof-Proof™ simplifies setup and improves reproducibility across both research and diagnostic workflows. Whether you’re running high-throughput assays or targeted qPCR panels, Goof-Proof™ offers the flexibility and precision you need without the interference of passive dyes.
Note: We also offer Goof-Proof™ Universal Probe Master Mix (Catalog # G-710) without ROX for instruments that do not require the ROX reference dye.
Goof-Proof™ Universal Probe Master Mix with ROX is a high-performance 2X master mix for fluorescent probe-based PCR applications such as SNP genotyping and quantitation. GoldBio HotStart Taq DNA Polymerase and dNTPs in an optimized buffer suitable for high qPCR sensitivity and multiplex reactions. Primers, probe and template not included.

Goof-Proof™ Tracking Buffer -
The 40X Tracking Buffer contains an inert blue dye. Adding the tracking buffer to the DNA template makes it “Goof-Proof” to track which reactions have had template added to the PCR reactions. Alternatively, Goof-Proof™ Tracking Buffer can be added to the master mix to distinguish wells with reaction buffer from empty wells. Tracking Buffer is not required for the reaction to function, but is designed to help alleviate mistakes.

ROX Reference Dye -
ROX is necessary for accurate Ct determination from well to well with certain instruments (such as some ABI and Stratagene PCR instruments, refer to your instrument instructions to see if ROX is required). ROX may add noise to melt curve analysis, which could be mistaken for real peaks. In the case of unexpected peaks, un-check “ROX” in the “Passive Reference Dye” box in the system software to prevent data collection from the ROX fluorescence channel and re-analyze the data.

TaqMan® Probe–Based Real-Time PCR Detection

Purpose: To perform highly specific and quantitative detection of DNA targets using hydrolysis probe technology.

How It Works: The hot-start Taq polymerase cleaves TaqMan® or dual-labeled probes during amplification. This separates the reporter from the quencher, generating a fluorescence signal proportional to the amount of target DNA.

ROX-Normalized Quantification Across qPCR Platforms

Purpose: To ensure consistent quantification on instruments requiring passive reference dye normalization.

How It Works: The ROX dye included in the mix acts as a stable internal reference, correcting for pipetting variation and instrument-related fluorescence fluctuations.
