Description GoldBio’s HEPES, Free Acid is a zwitterionic biological buffer widely used in cell culture, biochemical assays, molecular biology, and protein research applications.
HEPES is particularly valued for its ability to maintain physiological pH in the range of approximately 6.8 to 8.2 while exhibiting relatively low interference with many biochemical reactions and cellular processes. Unlike bicarbonate-based buffering systems, HEPES provides effective buffering capacity independent of carbon dioxide equilibrium, making it useful in open-air experimental systems and microscopy applications. HEPES is frequently incorporated into mammalian cell culture media, imaging buffers, electrophysiology solutions, and protein purification systems where stable pH control is critical for maintaining biological activity and cellular viability. The buffer demonstrates relatively low membrane permeability and is well tolerated in many biological systems when used at standard experimental concentrations. Because HEPES maintains buffering capacity near physiological pH and exhibits compatibility with a broad range of enzymes and biomolecules, it is commonly used in biochemical assays, diagnostic reagent preparation, and molecular biology protocols requiring reproducible pH stabilization.
HEPES free acid can be mixed with HEPES sodium salt to achieve the desired pH. Alternatively, HEPES free acid can be titrated with NaOH or KOH to achieve the desired pH.
