Protease Inhibitor Cocktails

ProBlock™ Gold 2D is a protease inhibitor cocktail specifically developed for sample preparation for IEF/2D- studies. No EDTA is used, which allows optimal action of nuclease activity for removing nucleic acids from the samples.ProBlock™ Gold 2D contains both irreversible and reversible protease inhibitors and inhibits serine, cysteine and metalloproteases etc. Due to the optimized concentration of the various inhibitors, the ProBlock™ Gold 2D shows excellent inhibition of protease activities and is therefore suitable for the protection of proteins extracted from animal cells / tissues, plants, yeast and bacteria etc. ProBlock™ Gold 2D at 1X concentration in extraction buffer at pH 7-8 inhibits over 90% of protease activities.

A broad range, 100X concentrated, ready-to-use protease inhibitor cocktail. ProBlock™ Gold Bacterial 2D contains optimized concentrations of various bacterial protease inhibitors, which provide excellent inhibition of protease activities during recombinant protein purification from bacteria. The cocktail contains both reversible and irreversible protease inhibitors to inhibit metallo, serine and cysteine proteases. It also contains specific inhibitors for bacterial proteases, such as aspartic proteases and aminopeptidases. A 1X concentration of Bacterial ProteaseArrest™ inhibits over 90% of protease activities.

ProBlock™ Gold Bacterial 2D cocktail does not use EDTA as its metalloprotease inhibitor as it would inhibit the activity of proteins that require divalent cations (Ca2+, Mg2+, or Mn2+) for their biological activity. In addition, EDTA would inhibit the purification of proteins using immobilized metal affinity chromatography (IMAC), for example His tagged or CBP tagged proteins. ProBlock™ Gold Bacterial 2Dcocktail is compatible with immobilized metal affinity chromatography.

ProBlock™ Gold Bacterial is supplied with additional, optional EDTA and 1,10-Phenanthroline monohydrate which are provided for optimal inhibition of metalloproteases and may be added in the extraction buffer or lysate as needed.

GoldBio's ProBlock™ Gold Extra Strength Protease Inhibitor Cocktail is a concentrated, broad-spectrum inhibitor formulation designed for demanding protein extraction and purification applications where increased protease protection is needed.

It contains 50% higher levels of protease inhibitors than the standard ProBlock™ Gold formulation and combines reversible and irreversible inhibitors targeting serine, cysteine, aspartic, aminopeptidase, and other protease activity.

At a 1X concentration in extraction buffer at pH 7 to 8, ProBlock™ Gold Extra Strength inhibits more than 90% of protease activity in GoldBio's mouse pancreas extract test system.

The disclosed formulation contains AEBSF, aprotinin, bestatin, E-64, leupeptin, pepstatin A, and PMSF. Combining inhibitors with different target specificities provides broader protection than relying on a single protease inhibitor.

The Extra Strength formulation is especially useful for samples with high endogenous protease activity or when maximizing recovery of intact proteins during initial extraction and purification is important.

ProBlock™ Gold Extra Strength is supplied with separate EDTA and 1,10-phenanthroline monohydrate solutions for optional metalloprotease inhibition. Researchers can add either chelator when additional metalloprotease protection is needed or leave them out when metal chelation could interfere with the experiment.

This is particularly important for proteins that require Ca²⁺, Mg²⁺, or Mn²⁺ for biological activity and for immobilized metal affinity chromatography. Omitting EDTA and 1,10-phenanthroline can also improve compatibility with applications such as isoelectric focusing.

Mechanism

ProBlock™ Gold Extra Strength uses a multi-inhibitor approach to provide increased protection against proteolysis in samples with elevated or difficult-to-control protease activity. Its higher inhibitor content provides increased inhibitor availability during protein extraction and purification from samples with substantial protease activity. The combination targets multiple protease classes simultaneously, reducing the likelihood that active endogenous proteases will continue degrading proteins after cells or tissues are disrupted.

ProBlock™ Gold mammalian protease inhibitor cocktail contains optimized concentrations of various mammalian protease inhibitors, which provide excellent inhibition of protease activities during protein purification from mammalian cells and tissues. The cocktail contains both reversible and irreversible protease inhibitors to inhibit serine, cysteine and specific inhibitors for mammalian proteases, such as aminopeptidases. A 1X concentration of ProBlock™ Gold Mammalian inhibits over 90% of protease activities.

The cocktail contains both reversible and irreversible protease inhibitors to inhibit serine, cysteine, metalloproteases and specific inhibitors for plant proteases, such as aspartic proteases and aminopeptidases. A 1X concentration oft ProBlock™ Gold Plant inhibits over 90% of protease activities.

ProBlock™ Gold Protease Inhibitor Cocktail [100X] is a ready-to-use liquid cocktail for protecting proteins from degradation during cell and tissue extraction, lysis, and protein purification.

The broad-spectrum formulation combines reversible and irreversible inhibitors targeting serine, cysteine, and other proteases. It is suitable for protein isolation from animal and plant tissues, yeast, and bacteria.

At a 1X concentration in extraction buffer at pH 7 to 8, ProBlock™ Gold inhibits more than 90% of protease activity in GoldBio's mouse pancreas extract test system  (e.g. Mouse Pancreas Extract, 0.5 mg/ml protein).

The 100X liquid format allows you to pipette the amount needed directly into an extraction buffer or lysate without dissolving tablets or preparing individual inhibitor stocks.

Its formulation contains AEBSF, aprotinin, bestatin, E-64, leupeptin, pepstatin A, and PMSF. Together, these inhibitors provide broader protease coverage than a single inhibitor and help preserve intact proteins for purification and downstream analysis.

ProBlock™ Gold is supplied without EDTA in the primary cocktail, with a separate bottle of EDTA included for optional metalloprotease inhibition. This gives you control over when chelation is appropriate. EDTA can interfere with proteins that depend on Ca²⁺, Mg²⁺, or Mn²⁺ and is incompatible with immobilized metal affinity chromatography unless removed before purification.

Leaving EDTA out also makes the primary cocktail suitable for applications such as isoelectric focusing. When metalloprotease inhibition is needed, the supplied EDTA solution can be added separately to the extraction buffer or lysate.

ProBlock™ Gold Yeast/Fungi protease inhibitor cocktail contains optimized concentrations of various yeast and fungal protease inhibitors, which provide excellent inhibition of protease activities during protein purification from yeast or fungi. The cocktail contains both reversible and irreversible protease inhibitors to inhibit serine, cysteine, metalloproteases and specific inhibitors for yeast or fungal proteases, such as aspartic proteases. At 1X concentration, ProBlock™ Gold Yeast/Fungi inhibits over 90% of protease activities.
