    Sulfadiazine is used to suppress susceptible Gram-positive and Gram-negative bacteria and to investigate folate biosynthesis and sulfonamide resistance mechanisms. It is commonly incorporated into agar or broth to apply selective pressure in strains carrying resistance determinants or to evaluate mutations affecting bacterial metabolic pathways. Sulfadiazine belongs to the sulfonamide class of antimicrobials and functions as a structural analog of para-aminobenzoic acid, a substrate required for folate synthesis in bacteria.

    Mechanism: Sulfadiazine competitively inhibits dihydropteroate synthase, the enzyme responsible for incorporating para-aminobenzoic acid into dihydropteroate during tetrahydrofolate synthesis. By disrupting folate production, it limits the availability of essential cofactors required for nucleotide biosynthesis, ultimately impairing DNA replication and cell division in susceptible organisms. This targeted metabolic interference makes sulfadiazine useful for examining folate pathway function, sul gene mediated resistance, and bacterial growth under defined nutritional conditions.
