TrueBlack™ is a new alternative to Sudan Black B for the quenching of lipofuscin autofluorescence in tissue sections for immunofluorescence staining. Lipofuscin consists of highly autofluorescent granules of oxidized proteins and lipids that build up in the lysosomes of aging cells. Lipofuscin granules fluoresce brightly in all channels used for fluorescence microscopy and accumulate in a variety of cell and tissue types with age. It’s necessary to quench or mask lipofuscin fluorescence in certain adult human tissues or aged animal tissues in order to properly image specific immunofluorescence signals.
Sudan Black B has traditionally been used to quench lipofuscin autofluorescence by incubating tissue sections with the dye after immunofluorescence staining. However it also introduces uniform, non-specific background fluorescence in the red and far-red channels, limiting the use of fluorescent dyes in those wavelengths.

Trypsin is a serine protease found in the digestive system of many vertebrates where it hydrolyzes proteins. Through trypsinisation, it cleaves peptides at the carboxyl side of arginine and lysine, unless they are followed by a proline.
Trypsin has an optimal operating pH of about 7.5-8.5 and optimal operating temperature of about 37°C. Unlike chymotrypsin, the activity of trypsin is not affected by the inhibitor tosyl phenylalanyl chloromethyl ketone (TPCK).

Trypsin inhibitor from soybeans is a monomeric protein containing 181 amino acid residues in a single polypeptide chain cross-linked by two disulfide bridges. Soybean trypsin inhibitor forms a 1:1 complex with the active protease site. Inhibition may be reversed and is pH dependent. The optimal pH for trypsin binding is 8.0.

Tryptophol is an aromatic alcohol found as a secondary product of alcoholic fermentation that induces sleep in humans. Typically synthesized in yeasts it is also produced by the trypanosomal parasite Trypanosoma brucei resulting in sleeping sickness. Tryptophol is a possible functional analog of serotonin or melatonin and shows genotoxicity in vitro. Saccharomyces cerevisiae uses it as a quorum sensing molecule.
Tryptophol is used as a precursor to the synthesis of tryptamines like DMT and the drug indoramin along with TNF-alpha modulators. When administered to rodents it resulted in significantly depressed antibody production.

Thymic stromal lymphopoietin (TSLP) is a hemopoietic protein belonging to the cytokine family and is known to play an important role in the maturation of T cell populations through activation of antigen presenting cells. It is mainly expressed in tissues, including heart, liver and prostate. TSLP signals through a heterodimeric receptor complex composed of the thymic stromal lymphopoietin receptor and the IL7R alpha chain. Like IL7, TSLP induces phosphorylation of STAT3 and STAT5, but uses kinases other than the JAKs for activation. After binding STAT5, phosphorylation is induced which results in the expression of downstream transcription factors. TSLP enhances the maturation of CD11c+ dendritic cells and induces allergic inflammation by directly activating mast cells. TSLP’s expression is linked to many disease states including asthma, inflammatory arthritis, atopic dermatitis, and eczema and other allergic states but the factors which induce the activation of TSLP release are not clearly defined. Human TSLP shares approximately 43 % amino acid sequence identity with mouse TSLP.
