The pACYC plasmid series is another set of commonly used cloning vectors in molecular biology and biotechnology. PACYC184 has a p15A origin of replication with chloramphenicol and tetracycline resistance genes. Unlike the pUC or pBR325 vectors, pACYC184 plasmids are characterized by a low copy number. This means fewer copies of the plasmid are present in each bacterial cell, which can be advantageous for cloning and expressing genes that are toxic to the host cells when present in high quantities. pACYC184 has a small size of 4245 bp and is suitable for cloning small to medium-sized DNA fragments. PACYC plasmids are especially useful in experiments where multiple plasmids are required within the same cell, or when cloning genes that are deleterious to host cells at high copy numbers.

The Yeast Lysis Buffer Kit is useful for extraction of soluble proteins from yeast cells. It is a proprietary improvement on the Zymolyase® based spheroplast preparation and extraction of soluble proteins from yeast cells.

This kit is provided with an optional protocol to make spheroplast and remove lytic enzyme Zymolyase®, prior to lysis and extraction of yeast proteins. Additionally, it has been used to develop a method used to extract DNA from Cryptosporidium oocysts. It may be used as a substrate for the development of DNA detection methods.

CHES is a zwitterionic buffering agent used in biochemistry and molecular biology. It is a cyclohexylamine buffer that is useful for a pH range of 8.6 – 10.0 and commonly used to study enzymatic processes above physiological pH. CHES displays only weak or negligible complexation capabilities for some metals making it a suitable non-coordinating buffer for use in most solutions containing metal ions. CHES displays high affinity for the iodoacetate binding site of liver alcohol dehydrogenase, protecting the enzyme from inactivation by iodoacetic acid.
