    Carbenicillin is a member of the penicillin β-lactam antibiotics. However, unlike most β-lactams, carbenicillin disodium is limited to primarily gram-negative bacteria including Pseudomonas aeruginosa and common enteric species. It can be used in selection protocols and is often preferred over ampicillin due to its increased stability. This allows for less contamination of unwanted bacterial colonies. In addition, carbenicillin has been used to regulate bacterial growth in plants, yielding low regeneration frequencies.     Proteinase K is a highly reactive nonspecific serine protease that belongs to the subtilisin family of proteins. It cleaves at the carboxylic acid side of aliphatic, aromatic, or hydrophobic amino acids. Proteinase K is capable of inactivating RNases and DNases and is used in the isolation or preparation of high molecular weight nucleic acids. Proteinase K is also useful for helping to characterize enzymes, due to its cleavage specificity. This enzyme was designated proteinase K because of its ability to hydrolyze keratin. Proteinase K is stable in a wide variety of detergents and buffer salts and at various temperatures and pH. The isoelectric point of proteinase K is 8.9.

    5-Bromo-4-chloro-3-indolyl β-D-galactopyranoside is most commonly known as X-Gal. X-gal is a histochemical substrate for β-galactosidase which cleaves X-Gal to yield an insoluble blue precipitate.

    X-Gal is a mainstay in molecular biology cloning applications in which it is used to detect the activity of β-galactosidase. X-Gal is used to detect the insertion of foreign DNA into the lacZ region of a plasmid DNA. Insertion of DNA into the lacZ region results in the loss of β-galactosidase activity. Bacteria cells that retain active β-galactosidase will result in characteristic blue colonies. Successful disruption of the lacZα gene disrupts the α-complementation of the β-galactosidase gene and the precipitate does not form, resulting in white colonies.

    Many other applications also use X-Gal as a substrate to detect β-galactosidase activity. These include β-galactosidase -antibody linked immunoassays and immunohistochemistry, coliphage detection based on β-galactosidase induction and the detection of micrometastasis formation during tumor progession.

    Lysozyme is an enzyme capable of breaking down the bacterial cell wall, and is typically used to enhance protein extractions or nucleic acid extractions. It works by hydrolyzing the 1,4-β-linkages located between the N-acetylmuramic acid and the N-acetyl-D-glucosamine residues in peptidoglycan.

    Gram-positive cells have an increased susceptibility to the actions of lysozyme since gram-positive cells have a higher quantity of peptidoglycan in the cell wall. Gram-negative cells are less vulnerable, but can still be hydrolyzed in the presence of EDTA.

    Nickel-NTA Agarose Resin has a binding capacity of ~50 mg/ml. In addition, it allows for purification of proteins under native or denaturing conditions. GoldBio Nickel Agarose works very well with the His-Tag Buffer Set.

    NTA cross-linked Agarose resin consists of nitrilotriacetic acid groups ligated by stable ether linkages via a spacer arm. NTA is a tetravalent chelating agent, covalently coupled to cross-linked agarose beads, providing a higher specificity and lower ion leaching than IDA linked resins. NTA resins have also been shown to be more robust in the presence of higher concentrations of EDTA, but may require a higher imidazole concentration for protein elution. This resin is loaded with Ni 2+. The resulting, ready-to-use resin is ideal for rapid purifications of His-tagged proteins.
