    Demeclocycline hydrochloride is used to inhibit susceptible Gram-positive and Gram-negative bacteria and to study protein synthesis inhibition and tetracycline resistance mechanisms.

    It is commonly incorporated into culture media or susceptibility assays to suppress sensitive organisms and to evaluate ribosomal targeting antibiotics in comparative studies.

    Demeclocycline is a member of the tetracycline class that binds reversibly to the 30S ribosomal subunit, blocking the attachment of aminoacyl tRNA to the A-site of the ribosome. This action prevents elongation of the growing polypeptide chain and inhibits bacterial protein synthesis in susceptible strains. Its well characterized ribosomal mechanism makes it useful for examining translational control, resistance determinants such as efflux pumps and ribosomal protection proteins, and selective growth conditions in microbiological systems.
